ABSTRACT

Nonalcoholic fatty liver disease (NAFLD), characterized by hepatosteatosis and steatohepatitis, is intrinsically related to obesity. Our previous study reported on the anti-obese activity of α,β-amyrin (AMY), a pentacyclic triterpene isolated from Protium heptaphyllum. This study investigated its ability to prevent fatty liver and the underlying mechanism using the mouse model of NAFLD. NAFLD was induced in male Swiss mice fed a high fat diet (HFD) for 15 weeks. The controls were fed a normal chow diet (ND). The mice were simultaneously treated with AMY at 10 and 20 mg/kg or fenofibrate at 50 mg/kg. Lipid levels along with metabolic and inflammatory parameters were assessed in liver and serum. The liver sections were histologically examined using H&E staining. RT-qPCR and western blotting assays were performed to analyze signaling mechanisms. Mice fed HFD developed severe hepatic steatosis with elevated triglycerides and lipid droplets compared with ND controls. This was associated with a decrease in AMP-activated protein kinase (AMPK) activity, an increase of mechanistic target of rapamycin complex 1 (mTORC1) signaling, and enhanced sterol regulatory element binding protein 1 (SREBP1) expression, which have roles in lipogenesis, inhibition of lipolysis, and inflammatory response. AMY treatment reversed these signaling activities and decreased the severity of hepatic steatosis and inflammatory response, evidenced by serum and liver parameters as well as histological findings. AMY-induced reduction in hepatic steatosis seemed to involve AMPK-mTORC1-SREBP1 signaling pathways, which supported its beneficial role in the prevention and treatment of NAFLD.

ABSTRACT

This study investigated the acute blockade of endogenous melatonin (MLT) using Luzindole with or without systemic lipopolysaccharide (LPS) challenge and evaluated changes in inflammatory and oxidative stress markers in the mouse jejunum. Luzindole is an MT1/MT2 MLT receptor antagonist. Both receptors occur in the small intestine. Swiss mice were treated with either saline (0.35 mg/kg, ip), Luzindole (0.35 mg/kg, ip), LPS (1.25 mg/kg, ip), or Luzindole+LPS (0.35 and 1.25 mg/kg, ip, respectively). Jejunum samples were evaluated regarding intestinal morphometry, histopathological crypt scoring, and PAS-positive villus goblet cell counting. Inflammatory Iba-1, interleukin (IL)-1β, tumor necrosis factor (TNF)-α, nuclear factor (NF)-kB, myeloperoxidase (MPO), and oxidative stress (NP-SHs, catalase, MDA, nitrate/nitrite) markers were assessed. Mice treated with Luzindole, LPS, and Luzindole+LPS showed villus height shortening. Crypt damage was worse in the LPS group. Luzindole, LPS, and Luzindole+LPS reduced the PAS-goblet cell labeling and increased Iba-1-immunolabelled cells compared to the saline group. Immunoblotting for IL-1β, TNF-α, and NF-kB was greater in the Luzindole group. The LPS-challenged group showed higher MPO activity than the saline and Luzindole groups. Catalase was reduced in the Luzindole and Luzindole+LPS groups compared to saline. The Luzindole group showed an increase in NP-SHs, an effect related to compensatory GSH activity. The acute blockade of endogenous MLT with Luzindole induced early changes in inflammatory markers with altered intestinal morphology. The other non-detectable deleterious effects of Luzindole may be balanced by the unopposed direct action of MLT in immune cells bypassing the MT1/MT2 receptors.

All mice were euthanized 60 days after infection. Praziquantel was the positive control in the experiment. Doses of 200 mg/kg (Mv-EO) and ROT (141.9 mg/Kg) resulted in a significant reduction in fluke burden (72.44% and 74.48%, respectively). There was also marked reduction in liver, intestinal and faecal and changed oogram pattern, compared to infected untreated mice. Considering the results obtained, further biological studies are required in order to elucidate the mechanism of schistosomicidal action on against adult S. mansoni.


Resumo O tratamento da esquistossomose é dependente de uma única droga, praziquantel (PZQ). O desenvolvimento da resistência de PZQ tem atraído atenção de muitos pesquisadores por medicamentos alternativos. Um tratamento viável e promissor é o estudo das plantas medicinais como uma nova abordagem para o tratamento experimental para esquistossomose. O presente trabalho objetivou avaliar a atividade esquistossomicida in vivo óleo essencial de Mentha x villosa (OE-Mv) e rotundifolona (ROT) contra Schistosoma mansoni. Foram utilizados camundongos Swiss webster (Mus musculus) fêmea de trinta dias de idade pesando 50 gramas. Os camundongos foram infectados com 80 cercárias de S. mansoni (cepa BH) e administrado por via oral OE-Mv (50, 100 e 200 mg/Kg) e ROT (35,9, 70,9 e 141,9 mg/Kg) apos 45 dias de infecção durante 5 dias consecutivos. Todos os animais foram eutanasiados 60 dias após a infecção. Praziquantel foi o controle positivo no experimento. O tratamento dos camundongos infectados com doses de 200 mg/kg (OE-Mv) e rotundifolona (141,9 mg/Kg) resultaram em redução significativa dos vermes (72.44% e 74.48%, respectivamente). Foi observado também redução no fígado, intestino e fecal e alteração no padrão do oograma, em comparação aos camundongos infectados e não tratados. Considerando os resultados obtidos, mais estudos biológicos são necessários a fim de elucidar o mecanismo de ação esquistossomicida contra adultos de S. mansoni.

ABSTRACT

Lymph node metastases are an independent prognosis factor in gastric carcinoma (GC) patients. Radical lymphadenectomy can improve survival but it can also increase surgical morbidity. As a principle, sentinel node (SN) navigation surgery can avoid unnecessary lymphadenectomy without compromising prognosis. In this pilot study, 24 patients with untreated GC were initially screened for SN navigation surgery, of which 12 were eligible. Five patients had T2 tumors, 5 had T3 tumors and 2 had T1 tumors. In 33% of cases, tumor diameter was greater than 5.0 cm. Three hundred and eighty-seven lymph nodes were excised with a median of 32.3 per patient. The SN navigation surgery was feasible in all patients, with a median of 4.5 SNs per patient. The detection success rate was 100%. All the SNs were located in N1 and N2 nodal level. In 70.9% of cases, the SNs were located at lymphatic chains 6 and 7. The SN sensitivity for nodal staging was 91.6%, with 8.3% of false negative. In 4 patients who were initially staged as N0, the SNs were submitted to multisection analyses and immunohistochemistry, confirming the N0 stage, without micrometastases. In one case initially staged as negative for nodal metastases based on SN analyses, metastases in lymph nodes other than SN were found, resulting in a 20% skip metastases incidence. This surgery is a reproducible procedure with 100% detection rate of SN. Tumor size, GC location and obesity were factors that imposed some limitations regarding SN identification. Results from nodal multisection histology and immunohistochemistry analysis did not change initial nodal staging.

ABSTRACT

Avaliou-se a suplementação de dietas com fósforo disponível (Pd) para suínos selecionados para deposição de carne, utilizando-se 100 leitões com média de peso inicial de 15,03±0,33kg, distribuídos em delineamento de blocos ao acaso, com cinco tratamentos, cinco repetições e quatro animais por unidade experimental. Os tratamentos foram dieta basal sem suplementação de fosfato bicálcico e outras quatro dietas obtidas pela suplementação da dieta basal com fosfato bicálcico, resultando em cinco concentrações de Pd, 0,106; 0,221; 0,310; 0,406; e 0,493%. As porcentagens de Pd influenciaram o consumo diário de ração, que aumentou de forma linear. O ganho de peso diário aumentou e a conversão alimentar melhorou de forma quadrática até a suplementação máxima de 0,450 e 0,390% de Pd, respectivamente. Não houve efeito da suplementação das dietas sobre a resistência óssea. As porcentagens de Pd influenciaram de forma quadrática os teores de cálcio e de fósforo nos ossos, que aumentaram até a adição máxima de 0,388 e 0,369% de Pd, respectivamente. Não houve efeito da porcentagem de Pd sobre o teor de cinza óssea. Suínos dos 15 aos 30kg exigem maior concentração de Pd para máximo ganho de peso (0,450% ou 5,33g/dia) e melhor conversão alimentar (0,390% ou 4,52g/dia) em relação às recomendações atuais.


The supplementation of diets with available phosphorus (aP) for pigs selected for meat deposition was evaluated, using 100 commercial hybrid piglets, 50 castrated males and 50 females, with average initial weight of 15.03±0.33kg allotted in a randomized block design with five treatments, five replicates, and four pigs, 2 males and 2 females per experimental unit. The treatments were a basal diet and other four diets obtained through basal diet supplementation with dicalcium phosphate to obtain five aP concentrations, 0.106, 0.221, 0.310, 0,406, and 0.493%. The percentages of aP influenced daily feed intake, which increased linearly. Daily weight gain increased and feed conversion improved quadratically up to 0.450 and 0.390% maximum responses. There was no effect of diet supplementation on bone strength. The percentages of aP influenced the calcium and phosphorus contents in the bones, which increased quadratically up to 0.388 and 0.369% maximum responses, respectively. There was no effect of aP percentage in the bone ashe content. Pigs from 15 to 30kg require greater amount of aP in the diet for maximal weight gain (0.450% or 5.33g/d) and greatest feed conversion (0.390% or 4.52g/d) compared to current requirements.

Leiomiomas son neoplasias benignas de músculo liso, poço frecuentes en cavidad bucal, que presentan crecimiento lento y se asemejan a otras lesiones de tejido conectivo. Clinicamente de tamaño entre 0,5 y 3 cm de diámetro, los leiomiomas vasculares generalmente tienen base pedunculada y coloración diversa debido a su ubicación y vascularización. El propósito de este artículo es presentar un reporte de caso de leiomioma vascular en lábio inferior y discutir su etiología, diagnóstico diferencial y tratamiento


Leiomyomas are benign smooth muscle neoplasms, rarely seen in oral cavity, that presents slow growth and are similar to others lesions of conjuntive tissue. Clinically with a size between 0,5 and 3 cm in diameter, vascular leiomyomas generally have pedunculated basis and diverse coloration due to location and vascularity. The purpose of this article is to present a case report of oral vascular leiomyoma in the lower lip and discuss their etiology, differential diagnosis and treatment

ABSTRACT

Cinco vacas holandesas pluríparas, com cânulas no rúmen e no duodeno, foram distribuídas em delineamento de quadrado latino 5 x 5. As vacas foram submetidas a cinco rações experimentais contendo 40 por cento de cana-de-açúcar, 60 por cento de concentrado e cerca de 30 por cento de amido. As rações diferiram quanto ao processamento ou à fonte principal do amido utilizado: milho grosseiramente moído, milho finamente moído, milho floculado a 310g/l, milho floculado a 360g/l ou raspa de mandioca. Não houve diferença (P>0,05) no consumo de matéria seca entre os tratamentos. A digestibilidade ruminal do amido foi maior na ração que continha raspa de mandioca. As digestibilidades ruminal da fibra em detergente neutro e da fibra em detergente ácido não diferiram entre os tratamentos. Os dados médios de pH ruminal se mantiveram acima de 6,0, exceto às 2 e às 4h após a alimentação com a dieta que continha raspa de mandioca. Não houve efeito significativo dos tratamentos sobre a concentração de ácidos graxos voláteis totais. O processo de floculação promoveu aumento da digestibilidade do amido do milho, em relação à moagem de forma grosseira. A digestibilidade ruminal do amido presente na raspa de mandioca foi maior do que a do milho, independentemente da forma de processamento utilizada.


Five multiparous lactating Holsteins cows, cannulated in the rumen and proximal duodenum, were used in a 5 x 5 latin square. Cows were fed a 40:60 forage: concentrate diet (40 percent fresh sugar cane and 60 percent concentrate). Diets were formulated to have 30 percent of starch and treatments were starch sources and ration processing forms: cracked corn, finely ground corn, flaked corn at 310g/l, flaked corn at 360g/l, or cassava scrapings. No difference (P>0.05) was observed among treatments for dry matter intake. Starch ruminal digestibility was higher for cassava scrapings treatment. NDF and ADF digestibility were similar among treatments. Average values of ruminal pH were above 6, except at 2 and 4h after feeding the cassava diet. There was no effect of treatment on concentrations of total volatile fatty acids. The flocculation process increased corn starch digestibility. Ruminal degradability of the cassava scrapings starch was higher than corn, without effect of processing form.

Eriocnema fulva Naudin is an endangered herbaceous, perennial, iteroparous, evergreen species geographically restricted to southeastern-center Minas Gerais State, SE Brazil. The individuals occur as patches on rocky riverbanks shaded by seasonal semideciduous Atlantic forest; they are fixed by roots and have a pending stem. Aiming to investigate leaf development and its importance for individual survival, fifteen contiguous plots (1 x 1 m) were set down in Jambreiro Forest (19° 58’-59’ S and 43° 52’-55’ W, 800-1100 m altitude), in the municipality of Nova Lima. A total of 260 individuals with the largest leaf blade length > 1 cm was tagged and measured in 1997, 1998, and 1999. Leaf expansion was recorded each month during 26 months until April 2000. Plant size was measured through leaf blade length, petiole length, stem length, and number of leaves. Significant changes were detected only after two years, thus indicating that plant growth is slow. The proportion of surviving leaves after two years was 60 percent. Total blade expansion took over 14 months, a slow growth rate when compared to leaves of other tropical forest canopy and understory species. Long leaf lifespans are to be found in plants exhibiting slow growth, and we observed that some leaves lived longer than three years. Petiole growth can help to better position the leaf in the search for light, thus contributing to the growth and survival of the plant. The relationships among size measures were significant, reinforcing the great contribution of leaf size for plant size. The age of the largest individual was estimated as 36 years based on the median annual leaf production rate.


Eriocnema fulva é uma espécie herbácea, perene, iteropárica, com distribuição geográfica restrita ao centro-sudeste do Estado de Minas Gerais, Brasil, e ameaçada de extinção. Os indivíduos ocorrem agrupados em paredões rochosos, na beira de riachos sombreados pela Mata Atlântica estacional semidecídua e têm caule que pendem do barranco. Com o objetivo de investigar o desenvolvimento da folha e sua importância para a sobrevivência dos indivíduos, foram instaladas 15 parcelas contíguas de 1 x 1 m na Mata do Jambreiro (19° 58’-59’S, 43° 52’-55’ W, 800-1100 m de altitude), município de Nova Lima. Todos os indivíduos cuja maior folha tivesse comprimento do limbo maior ou igual a 1 cm foram marcados, sendo medidos em 1997, 1998 e 1999. O tempo de expansão da folha foi registrado mensalmente durante 26 meses até abril de 2000. Na área amostrada foram marcados 260 indivíduos cujo limbo foliar tinha comprimento > 1cm. O tamanho das plantas, medido através do comprimento do limbo e do pecíolo e do número de folhas, apresentou mudanças significativas somente após dois anos, indicando que o crescimento é lento. A proporção das folhas sobreviventes após dois anos foi de 60 por cento. O tempo necessário para a expansão da folha foi superior a 14 meses e é lento quando comparado com o de outras espécies do dossel ou do sub-bosque da floresta tropical. Maior longevidade foliar tende a ser encontrada em plantas de crescimento lento. Observamos que algumas folhas tiveram maior longevidade que 3 anos. O crescimento do pecíolo pode ajudar no melhor posicionamento da folha na busca de luz no sub-bosque, contribuindo para o crescimento e sobrevivência da planta. As relações entre a folha e demais medidas de tamanho foram significativas, indicando sua grande importância para o tamanho de cada planta. A idade do indivíduo de maior tamanho foi estimada em 36 anos através da taxa mediana de produção anual de folhas.

Eriocnema fulva Naudin is an endangered perennial herbaceous plant, endemic to Minas Gerais state, Brazil. This study was conducted in the Jambreiro Forest (19° 58' -59'S and 43° 52' -55' W, 800-1100 m altitude). In an attempt to describe the population size structure and its association with individual fertility, fifteen 1 x 1 m contiguous plots were set. We tagged, counted, and measured a total of 260 individuals in 1997, 1998 and 1999. Young individuals with leaf lamina lengths < 3.4 cm comprised 33 percent of the total sampled, indicating that the population was reproducing locally. The number of leaves varied significantly, growth differences being detected only after two years of measurements. Stem length was the variable that best showed population size variation. The length of the largest leaf lamina was the best indication of its development phase. Assessing the number of leaves helped to evaluate the alteration in plant size during the study. The probability that individuals with laminas > 10 cm in length did not reproduce was 2.69 percent. The highest survival probability of the large-sized individuals confirmed the strong correlation between size and survival. The data indicated that size is important for the fertility of E. fulva, and it may be one of the relevant aspects to be considered for analyses of survival probability. The intraspecific competition, which was indicated by negative correlation between fruit production per size unit and density, can affect fertility, as larger plants had higher fruit production.

Este estudo foi realizado na Mata do Jambreiro (19° 58' -59' S e 43° 52'-55' W, 800-1100 m de altitude) Com o objetivo de descrever a estrutura de tamanho da população e suas relações com a fertilidade dos indivíduos, foram instaladas 15 parcelas contíguas de 1 x 1m. Marcamos, contamos e medimos um total de 260 indivíduos em 1997, 1998 e 1999. Os indivíduos jovens com o comprimento do limbo < 3,4 cm somaram 33 por cento do total amostrado, mostrando que a população estava reproduzindo-se no local. O número de folhas variou significativamente entre os anos, com diferenças de crescimento só detectadas após dois anos de medições. O comprimento do caule foi a variável que melhor mostrou a variação de tamanho na população. O comprimento da lâmina da maior folha foi a melhor expressão da fase de desenvolvimento da planta, e o número de folhas auxiliou na avaliação da alteração do tamanho ao longo do tempo. A probabilidade de indivíduos com limbos foliares > 10 cm não se reproduzirem foi de 2,69 por cento. A maior probabilidade de sobrevivência dos indivíduos de maior tamanho confirmou a forte correlação entre tamanho e sobrevivência. Os dados mostraram que o tamanho é importante para a fertilidade de Eriocnema fulva e pode ser relevante para análises de probabilidade de sobrevivência da espécie. A competição intra-específica, indicada pela correlação negativa entre a produção de frutos por unidade de tamanho e a densidade, pode influir na fertilidade, pois plantas maiores apresentaram maior produção de frutos.

ABSTRACT

E apresentado o caso de uma crianca do sexo feminino de 5 anos de idade que apresentava uma massa palpavel na fossa iliaca direita.Havia leucocitose com intensa eosinofilia (52%). Levada a cirurgia foi encontrada uma massa nodular na juncao ileo-cecal tendo sido ressecados em bloco o ileo terminal, ceco e apendice. O exame da peca cirurgica mostrava espessamento da parede do ceco com areas de necrose de permeio. Histologicamente havia denso infiltrado eosinofilico por toda a parede com alguns vasos arteriais da serosa e submucosa apresentando em seu lumen seccoes transversais de parasitos identificado como sendo Angiostrongylus costaricensis e outros exibindo trombose intravascular. O presente registro constitui o quinto caso brasileiro de parasitismo por esse angiostrongilideo

ABSTRACT

Descreve-se o caso de uma paciente de 12 anos de idade, portadora de insuficiencia mitral reumatica, que desenvolveu endocardite bacteriana por Staphylococcus aureus. A infeccao determinou a formacao de um aneurisma sacular do ventriculo esquerdo, chegando a romper-se no saco pericardico. Realizou-se uma operacao de emergencia na qual se substituiu a valva mitral e se reparou o aneurisma,mas a paciente faleceu devido a fibrinolise. A raridade e os principais aspectos clinicos e cirurgicos desta condicao sao discutidos

ABSTRACT

Vinte e nove casos infectados de retarde de consolidacao,pseudartrose e perda de substancia foram tratados por periodo medio de 20 meses. Varios procedimentos foram empregados com predominancia da fixacao externa, osteossintese intramedular fechada com Kuntscher-Kaessman e tibializacao da fibula. Os resultados obtidos quanto a obtencao da consolidacao (79%) e cura da infeccao (89%) foram satisfatorios
